Tinctures give a temporary (30 seconds) moderate stat alteration to the user. They are tradable and dropped in purple bags from various enemies throughout the game including Greater Nature Sprites, Sarcophagi, Haunted Spirits, and Large Ghosts.
Effusions give a temporary (30 seconds) heavy stat bonus to everyone within a range of 6. Though formerly sold in the Nexus for Realm Gold, they are no longer able to be purchased in this way.
Purchasing chests in the Vault using Realm Gold will reveal a random Nexus item inside it, which can include either a tincture or effusion. Another way to obtain tinctures and effusions is by finding them as a reward in the Arena. Effusions can also be found in some mystery boxes.
The Treasure Pots from the Lost Halls and the Court of Oryx dungeons also have a chance of dropping both tinctures and effusions.
Every time you use a tincture or effusion that temporarily increases a stat, any other boost of the same stat used before the time limit (30 seconds) of the first item will only give half of the stat efficiency of the previous item’s efficiency; starting at 100%, then 50%, then 25%, etc. The stat increase itself is rounded down. The time limit for each use is still separate.
For example, if you use a tincture of defense your defense stat will boost to +15. If you use another one before waiting 30 seconds, the tincture will boost to +7, giving a total of +22.

In Release X32.4.1, tinctures and effusions received several updates:
Tinctures and effusions for the four remaining stats (attack, speed, vitality, wisdom) were added as drops from Xolotl the Lightning God, the Royal Cnidarian, Shaitan the Advisor, and The Puppet Master (Encore), respectively.
Tinctures/effusions of life/mana received new sprites (to match the color schemes of Health and Magic Potions, respectively), while the newly added tinctures/effusions of vitality/wisdom received the old sprites of life/mana, respectively.
Effusions were made non-soulbound, meaning they are now tradeable.
Effusions now drop from purple bags (previously blue).
